Admittedly, it's almost impossible to pick just three. But if pressed, I might select the following. (Although I could probably change my mind tomorrow. And these are just off the top of my head. There are probably others I'll think of that are even better after I post this! And yes, I'm aware I left off Janis Joplin!)
-Gladys Knight
-Linda Ronstadt
-Marilyn McCoo
Honorable Mention:
-Karen Carpenter

This is a loaded question with no REAL ANSWER.
aflem, you have a super list but there are a few others:
- Barbra Streisand
- Tina Turner
- Whitney Houston

Those are good choices, too, although I have to admit, I've never quite thought of Streisand as a rock singer, although she did have a few Top 40 hits. But what an amazing voice!

For my three....Karen Carpenter (of course)
Connie Francis (35 hits in the Top 40)
Aretha Franklin (45 hits in the Top 40)

Aretha Franklin and then a bunch of others…
Elkie Brooks, Wanda Jackson, LaVern Baker…

Linda Rostadt and Karen Carpenter (if she had only had better songs to work with) would definitely be on my list as well.
To add three names that have not yet been mentioned:
Darlene Love
Ronnie Spector
Peggy March

Upon further reflection, here are another three outstanding female voices:
Cass Elliot
Shirley Alston Reeves (The Shirelles)
Brenda Reid (The Exciters)

Yes, she's thought more as a pop star than rock. My exact sentiments but I will never forget her performance of "You'll Never Walk Alone" which capped the 2001 Grammy Awards - WOW!! Lots of clapping. Bravado and kudos to her for her strength - she is a classic textbook case of paranoia/stage fright which is why she rarely does live performances. Super memorable performance and no one could have done it better.

Rindy Ross-Quaterflash

Nobody has remembered Etta James yet...

Dusty Springfield should figure ion this list

Interesting no one has mentioned Diana Ross. I thought she had a very distinctive voice, but I wouldn't call it one of the greatest - although there's no denying The Supremes remain one of the finest so-called "girl groups" of all time.

Lorraine Ellison ( Stay With Me ), just blows me away
Aretha
Gladys Kinight

What! ... No Janis? (Joplin, I mean.)
